If your players want to unlock incredible power that fundamentally alters the setting, you might as well give their self-imposed quest the gravitas it deserves.

Ah, so this is where we get to some of the fun from Magical Mystery Cure, and the unfinished spell Plot Coupon it had. I do hope player-Twilight's a little more thoughtful than canon-Twilight and doesn't read it aloud next to the Elements.
More or less, depending of the headcanon and the story, Twilight completing the spell made her ascend to alicornhood is like a powerful wizard ascending to godhood or at last demi-godhood... How common is for a player or a character that it happen?
In 4e, it's... "expected" isn't quite the right word, but it's exactly the sort of thing an Arcane Character might use to narratively justify their Epic Destiny.
"Unlock" -- considering in show that they literally did have to unlock a container to (forgive me) taste the rainbow... Will be somewhat interesting to see how we get from StarSwirls Unfinished Spell (I had to resist "symphony") to the rainbow container -- especially if it skips over the plunder vine/return elements to tree.
